    The Recombinant Human Interleukin-10/IL10 was generated by the expression of a DNA fragment encoding 19-178aa of human IL10 in E.coli. The full-length of mature protein reached up to 95% in purity determined by SDS-PAGE and HPLC analyses. It was also verified to be fully biologically active through its effect on a cell proliferation assay using murine MC/9-2 cells (ED50<1 ng/ml, specific activity >1.0x106 IU/mg). The endotoxin of this IL10 is less than 1.0 EU/μg as determined by the LAL method. Proteins in-stock are offered now. The IL10 proteins are devoid of any tag, but CUSABIO offers custom service for you to meet the need of specific tag(s). This recombinant IL10 protein may be used to produce anti-IL10 antibodies or in the immunology studies.

    IL10 is an immunosuppressive cytokine that plays a key role in regulating intestinal immune homeostasis. IL10 functions by binding to the IL10 receptor. The activation of STAT3 by IL10 is essential for the immunosuppressive effects of IL10. Animal studies have shown that IL10 controls the responses of effector Th1 and Th17 thus maintaining intestinal tolerance to microbiota. It has recently demonstrated that the failure of innate immune cells to respond to IL-10 is critically involved in the development of such T cell-driven intestinal inflammation. S. Veenbergen etc. demonstrated that the IL-10 signaling in dendritic cells controls IL-1β-mediated IFNγ secretion by human CD4+ T cells.

    Major immune regulatory cytokine that acts on many cells of the immune system where it has profound anti-inflammatory functions, limiting excessive tissue disruption caused by inflammation. Mechanistically, IL10 binds to its heterotetrameric receptor comprising IL10RA and IL10RB leading to JAK1 and STAT2-mediated phosphorylation of STAT3. In turn, STAT3 translocates to the nucleus where it drives expression of anti-inflammatory mediators. Targets antigen-presenting cells (APCs) such as macrophages and monocytes and inhibits their release of pro-inflammatory cytokines including granulocyte-macrophage colony-stimulating factor /GM-CSF, granulocyte colony-stimulating factor/G-CSF, IL-1 alpha, IL-1 beta, IL-6, IL-8 and TNF-alpha. Interferes also with antigen presentation by reducing the expression of MHC-class II and co-stimulatory molecules, thereby inhibiting their ability to induce T cell activation. In addition, controls the inflammatory response of macrophages by reprogramming essential metabolic pathways including mTOR signaling.
